Key instrumentation challenges in single-use chromatography

The biggest challenges in single-use chromatography are the measurement, control, and reliability issues introduced by disposable flow paths, plastic components, and changing assemblies between batches. Single-use chromatography removes cleaning and cross-contamination concerns, but transfers risk toward measurement confidence, automation repeatability, and process validation.

  • Reliable flow measurement in flexible disposable tubing: In chromatography, flow rate is one of the critical process variables. If the actual flow differs from the recipe, the entire separation mechanism changes. In single-use systems the piping geometry is not fixed, since tubing assemblies are replaced frequently, wall thickness and material may vary.
  • Pressure protection of expensive chromatography columns: A chromatography column is often the most expensive component in the purification train. The resin bed is designed to operate within a narrow pressure window. Disposable systems introduce additional uncertainty as flexible tubing expands, disposable manifolds create pressure losses. As a result, the pressure profile may look different after every setup.
  • Robust pH and conductivity measurements: Chromatography is fundamentally a chemistry-driven process. Small deviations may completely change protein binding behavior. Disposable sensors introduce uncertainty around drift, calibration stability and sensor ageing.
  • Maintaining calibration and GMP compliance with disposable sensors: With single-use every batch may use a different sensor, different tubing assembly or a different disposable manifold lot. This creates significant lifecycle-management complexity.
  • Repeatable control performance after every flow-path replacement: Single-use systems are inherently less repeatable than fixed installations. A control loop that was stable last month may suddenly become more sluggish because tubing length changed or air pockets are trapped differently.
